Reconstitution
|
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute in sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0mg/ml. Stock solutions should be apportioned into working aliquots and stored at ≤-20℃. Further dilutions should be made in appropriate buffered solutions.

Background
|
The TP53-inducible glycolysis and apoptosis regulator (TIGAR), also named fructose-2, 6-bisphosphatase TIGAR, is an enzyme that in humans is encoded by the C12orf5 gene. The protein functions by blocking glycolysis and directing the pathway into the pentose phosphate shunt. Expression of this protein also protects cells from DNA damaging reactive oxygen species and provides some protection from DNA damage-induced apoptosis. TIGAR activity can have multiple cellular effects. Recombinant human TIGAR expressed in E. coli is a 30.1kDa protein containing 270 amino-acid residues.

保存條件:
-20℃或更低溫度保存，至少一年有效。由于蛋白的每次凍融均會(huì)引起部分失活，所以首次配制成相應(yīng)濃度的儲(chǔ)存液后(請根據(jù)產(chǎn)品簡介中Reconstitution一欄的信息配制儲(chǔ)存液)，須分裝后-20℃或更低溫度凍存，以避免反復(fù)凍融。
注意事項(xiàng):
由于有些塑料管壁對某些蛋白有較強(qiáng)的吸附作用，溶液中的蛋白很容易粘附在管壁上，并且粘附后的蛋白很難與管壁分離。而載體蛋白(Carrier protein，如0.1% BSA等)的主要作用是預(yù)先封閉塑料管壁上的蛋白結(jié)合位點(diǎn)，使細(xì)胞因子或重組蛋白不會(huì)粘附于管壁。所以一定要使用產(chǎn)品簡介中Reconstitution一欄的信息配制儲(chǔ)存液。
